Low androgen status inhibits erectile function by inducing

The study found that the level of serum T and NO in the corpus cavernosum and the ratio of ICPmax/MAP in castration groups were
significantly lower than those in sham-operated groups or castration + T groups. This indicates that low androgen levels impair erectile function of rats. It was consistent with previous studies.6,22-24Compared with the sham-operated groups and castration + T replacement groups, the ratio of eNOS monomers/dimers in penile cavernous tissue of castration groups was significantly increased, and NO production of castration groups was significantly reduced. This indicates that eNOS uncoupling in the penile cavernous tissue of castrated rats was significantly increased. Therefore, the increased eNOS uncoupling under low androgens status may be one of the mechanisms for decreased ICPmax/MAP.
The mechanism of eNOS uncoupling is complicated. The decreased BH4/BH2 caused by the relative or absolute deficiency of BH4 may be the major cause of eNOS uncoupling.12,25 BH4 is synthesized by the de novo pathway and salvage pathway in cells. In the de novo pathway, BH4 is synthesized from guanosine 5-triphosphate (GTP) by the rate-limiting enzyme GTPCH1. In the salvage pathway with sepiapterin as the substrate, BH4 is synthesized from reduced BH2 by the rate-limiting enzyme DHFR. Meanwhile, BH4 is oxidized
